The South Western Highway between Bendall and Argyle roads in Argyle, about 10 kilometres north of country town Donnybrook, has been closed, with emergency crews on the scene.
A single car with a solo occupant is understood to be involved. The driver is believed to have non-life threatening injuries.
Police say the crash happened just before 5am.
